Yesterday I performed the serial dilution. There was significant growth in the initial broth cultures I prepared from all nine sample types. I think my lab techniques were organized and fairly efficient for the first time doing these dilutions. I plated each dilution starting from a factor of 1:10 down to 1;100,000. Today I finished the fixing protocol and will keep the samples stable in 95% ethanol to preserve the pellet and image them tomorrow. We must do this as not to completely dissolve them in tert-butanol because we no not have a running centrifuge at Fairchild to reform the pellet. I am hoping the pellet does not completely dissolve on it's own.
